Q: 32' brightness drop on single run. This is coming directly off 24v power supply to strip. Am I missing something?

Multiple parallel runs are preferred over long series runs. A power supply is needed at each amplifier on series runs. Voltage drop (and thus brightness drop) is significant over a long series run with one power supply.

Q: I have a 12v 5amp power supply connected, but it's not powering the amplifier. what are the power requirements if i have 4 16' rgbw strips connected?

You will need at least a 240 Watt power supply, which may be called a 20 Amp power supply. The amplifier needs to be connected to a control to turn it on though. You would connect one 16' strip of lighting to the control and power it by the 5 amp power supply and then go from that strip to the input on the amplifier. You could also use a 5 amp power supply and an amplifier for each 16' strip of lighting. Hope this helps.

You could yes. But they will all be dimmer than usual.

Depending on if you're using 12 or 24vdc RGB strips will dictate how many strips can be powered from one amplifier. Each amplifier should have a direct feed from the controller. I wouldn't jump from the end of a light strip to the next amp. Although it may work that way, I think there is more of a chance for potential issues down the road
